第五届CCTV相声大赛于2010年10月2日首播，每天有4个相声段子参加角逐，至10月8日结束。每个参赛节目的出场顺序都是由选手抽签决定的，每场比赛除参赛节目之外还有一个由历届相声大赛获大奖的演员或相声名家表演；连续6天的决赛结束后，10月8日晚还播出了相声大赛的颁奖晚会《今夜相声灿烂》。
